Relevance. Evaluation of the effect drugs on neurotransmitter processes is an important component pharmacodynamic studies. The quantitative determination monoamine neurotransmitters in brain structures laboratory animals urgent task pharmacology and physiology. Purpose study. Development a method for serotonin, dopamine, norepinephrine, histamine epinephrine rat homogenates using HPLC-MS/MS. Methods. isolation from rats was carried out by homogenizing biomaterial with acetonitrile hydrochloric acid. extraction purified liquid-liquid chloroform isopropanol. Monoamines were detected AB Sciex QTrap 3200MD mass spectrometer, chromatography performed Agilent Technologies 1260 Infinity II HPLC. Methanol deionized water used as eluent. Results. Sample preparation consisted centrifugation resulting homogenate, drying supernatant stream nitrogen, dissolution precipitate mobile phase, purification solution mixture An InfinityLab Poroshell 120 EC-C18 4.6×100 mm, 2.7 μm analytical column to separate neurotransmitters. total time chromatographic analysis 12 minutes, retention epinephrine, 2.8; 3.2; 5.4; 7.9; 2.2 respectively. range technique 25.0–5000.0 ng/g histamine, dopamine; 5.0–5000.0 serotonin 50.0–5000.0 norepinephrine. To test technique, we analyzed striatum intact Wistar rats. Conclusion. developed bioanalytical HPLC-MS/MS fully complies validation requirements. metrological characteristics make it possible estimate content high accuracy.
